Assessment of DNA extraction methods for detection of arbuscular mycorrhizal fungi in plant roots by nested-PCR Biological Sciences

DNA extraction methods were evaluated for the yield and purity of DNA recovered from mycorrhized roots and whether the recovered DNA is suitable for amplification of arbuscular mycorrhizal (AM) fungal SSU rDNA. The DNeasy Plant Mini Kit and three extraction buffers were used alone or in combination with either polyvinylpyrrolidone (PVP), polyvinylpolypyrrolidone (PVPP) and/or activated charcoal (AC). Among the extraction methods tested, those based on the CTAB buffers yielded more DNA than those based on the TE buffer and the DNeasy Plant Mini Kit. Rhizobial diversity from stem and root nodules of Discolobium and Aeschynomene Agronomy

Among the genera that exhibit stem nodulation, Aeschynomene and Discolobium are found in the Pantanal Mato-grossense (wetlands in the state of Mato Grosso, Brazil). Isolates obtained from the stem and root nodules of D. pulchellum, D. psoraleaefolium, D. leptophyllum and A. fluminensis were collected from various locations in the Pantanal de Poconé sub-region and phenotypically characterized and genotyped by restriction analysis of the 16S rDNA gene. Of the 282 isolates obtained from the stem and root nodules, 84.3% alkalized YMA media and 74.1% showed slow growth.
